I am afraid this is no option to change the hover color at the moment, however, you can use the CSS below by Adding it to the On-Page Css:
.wp-element-button:hover, .wp-block-button__link:hover {
background-color: #85fe6b;
}

I just now saw your addition about the plugin. This indeed works for the search bar, but not for the button to leave a comment “post comment”. I’ve tried with the css code in the page layout, but it doesn’t work. Do you have any other suggestions for this to adjust the hover color for the comment section?

On checking the post comments block as shown here : https://share.getcloudapp.com/jkuR7KjD. This is from a Gutenberg core block and not a Spectra block. I would request you contact the WordPress Gutenberg support team to help with this.
