I recommend using the My Account registration (or just turning on account creation on your checkout) for retail customers and keeping your wholesale registration separate.
The information you need to collect for wholesale is a lot more and different kinds of information than you would need from regular retail customers. No need to put those retail folks through the same process as that would hurt your conversion.

For your question regarding incentives for employees of your wholesale customers. What does the tier look like? Is it just an extra % off retail?
You could probably consider adding another “wholesale” role called “Wholesale Customer Employee” for example and anyone who qualifies for this can be assigned this role.

And for options around pricing for those people, in Prices Premium you can set pricing as a general % off site wide, % off per category, or set the actual pricing via fixed price or % off on the product level too. You can even do a mix of all these.
