Is there a way to create a view to display a single random testimonial every page load?

Yes. In the View editor, set order to random and quantity to one.

The plugin does not track which testimonials have already been displayed. It randomly selects one from the same list each time so you may see the same testimonial more than once before the entire list has been displayed.
