But if the file also gives a 404 when Autoptimize is disabled (by the parameter in the URL), then I’m afraid nothing in AO’s configuration can help here .. :-/

I don’t get a 404 when AO is disabled, only when it is enabled.

OK, do you still get the error when “aggregate JS” or even “optimize JS” in Autoptimize is off?
The only enabled option was Optimize JS Code. I’ve disabled that and the error still occurs. I did not delete cache when saving that change though, is that required?
deleting AO’s cache should not be needed, but you might have a page cache that is preventing you from seeing the change?
