Recently the Jetpack comments function has stopped working on our site. Regardless of which posting login is used, after clicking on the Submit button, the words “Submitting Comment” briefly appear and then it resolves as “404: File not found”.

Have you updated WordPress / Jetpack recently? Can you recall any changes made just before this happened?
In the first instance I would delete the Jetpack plugin then install a fresh copy.
