You can remove the MailPoet key from MailPoet > Settings > Key Activation and also ensure you reset your sending method settings in MailPoet > Settings > Send With…
If you remove the key and choose MailPoet Sending Service as your sending method, no emails will be sent.

I would prefer if I don’t have to change anything in the database (can be copied from live environment occasionally). Any possibility for a define or something?
What happens if we copy a database from live system where a key is specified (we will most likely get one soon after tests) to staging or local development environments? I guess it will not work (which is ok), but there are not problems/errors?

It may still work for a short time before the scheduled key check task runs (but it could be enough for some emails to get sent) so I recommend making the changes I described above right after your live site is copied to the staging environment.
