do I need API to manually translate?

my plugin has a po. file.
I scanned and obtained the words.
I’ve made manual translations.
I’ve saved the mo. file.

I see no changes, in the website, where I made changes.

Am I doing something wrong?
Not doing a step?
or do I need to turn on an API?
