Does this plugin work with BuddyPress?

WP-SpamShield protects the WordPress registration page. BuddyPress uses a different registration form that bypasses the default WordPress page, so no, as of right now it won’t work with the BuddyPress registration form. I’m not an expert on BuddyPress, so I’d have to look into it and see if it’s feasible. If so, that feature may be added at some point in the future. I hope that helps answer your question.
