Try opening the generated administrator/backups/perm.txt file , and look between lines 600 and 700, do you notice any large files that you could exclude?
Also, how much free disk space do you have available?

Something similar happened to me (I think). The plugin always worked just fine but this last time when I tried to backup, I got this:

UPDATE:
Changing from “AJAX Backup Refresh Mode” to “NORMAL Backup Refresh Mode” allowed for the backup to complete. No more errors.
Why is this?
(I still haven’t tested the backup to see if everything is ok)
The backup process fails due to the mysql server which drops the queries at some point with ” MySQL server has gone away ” error, i would suggest checking with your hosting support on this.

I was having this problem too, and seems to have solved it by reducing the “Files to process per request” to 30 and increasing the “Time between requests” to 2000. I still get the message every now and then with the main difference being that clicking the retry now works and the backup continues.
