Yes, it’s possible, but I haven’t yet realized it in Shortcode Builder. You can add the option exclude_posts manually in the shortcode, for example: exclude_posts=”41″, where 41 is ID of excluded post. Multiple posts must be separated by comma.
I’m going to add this feature in one of the following versions of the plugin.

Excluding posts by the tag does not work in this version of the plugin, but I will add this feature in the next version of the plugin.

I release the new version of the plugin with the ability to filter the posts by tags (only in a shortcode, in widgets this ability will be available in the next version). You can include or exclude post by a specific tag or multiple tags. Now only work with posts.

I have not tested this feature with WP 4.3, but as far as I know, in this update, there was nothing that would affect the operation of this feature.
I’ll check the compatibility and, if the case in WP 4.3 – try to fix the plugin.
