Smush optimizes your images in the gallery and replaces the original, non-optimized images with the optimized ones.
There shouldn’t be any issues with the process while Jetpack is active but if you do encounter any issues do let us know.

I’m afraid that to this kind of questions there’s no 100% sure answer to be given.
We didn’t have any reports about Smush and Jetapack having issues, but we don’t know which exactly module combination you have active on your site, which plugins and theme you using and which option, and if any of them can affect Jetpack, Smush or Smush running alongside Jetpack.
Smush plugin has over million active installations and I’m sure there’s a good number of them using it beside Jetpack 🙂
As I said, it should work just fine but saying 100% would mean there’s no chance of anything going wrong, which would be a false statement as it can depend on so many factors.
If you don’t want to take any chances best approach would be installing it first on a staging site until you’re sure that it suits your needs.
