About the profile page, do you have a link on your theme that takes to the backend profile? http://www.mysite.com/wp-admin/profile.
Normally we rewrite them to use the front end profile, let us know.

Sorry for my incorrect expression above, the wordpress default profile url is http://www.mysite.com/wp-admin/profile.php
In order to avoid misunderstandings each other, I would like show details:
For now, I am using Login with Ajax:https://ww.wp.xz.cn/plugins/login-with-ajax/, and It will show to user profile link which is wordpress default, please check at http://prntscr.com/hdylwa, once click “profile”, it does not redirect user to Userwp profile page, I mean it will still let user enter backend profile.
but I think all setting is right, please check my setting:http://prntscr.com/hdyng6

Thanks, I used the last one:
Multisite – Not network active – can be activated on a per site basis
In this mode, you activate the plugin in each site instead of activating network wide.
This works exactly like the single site.
In other words a separate UsersWP plugin related tables are created for each site. For other modes single tables are used.
Steps to activate:
Visit either subdomain.example.com/wp-admin/plugins.php or example.com/subfolder/wp-admin/plugins.php
and then click the “Activate” link
And post on your official support forum.
