You can close bookings for a day using the Scheduling Exceptions under Bookings > Settings > Schedule. You can learn more in the docs here:

I’m afraid it’s not yet possible to set a scheduling exception for a specific location.
I have the same problem,
I have some locations and each one has different scheduling but when I set them
only show the times scheduled in the main plug-in options.

Yes, the plugin doesn’t yet support different booking schedules for each location.
