The multisite will create a new site as either a subdomain or a subdirectory of the domain it is created on… You can then map that as you please using the multisite’s built-in domain mapping.
You may need to work with your DNS records and possibly your control panel to direct the new domain, subdomain, subdirectory, or subdirectory of a subdomain to work but the mapping in WordPress should be straight forward.
I’d recommend getting the main site to work properly then map in my most complicated subdomain with subdirectory next. Once you’ve got those two working well, then the rest is easy.

BTW: Don’t spend time looking at the WordPress file system for subdirectories either… subdirectories are not what you might think they are in WordPress.
Also: If you get default pages from the host then look at your CP to determine if the domain and or subdomain is being passed to WordPress properly.
