Must files exist?

We are migrating from a static html site to a wordpress. The new site has a completely different directory structure than the old site. I’d like to use the redirection plugin to create redirect rules for all of the old content without having to recreate the old directory structure and uploading all the old content. Can this be done?
The reason I’m asking is I read somewhere in the support docs a file (or directory) must exist before it can be redirected.
