Hi,

I’m trying to use this plugin with a wp multisite network. The main site (the first site it works well. All the files are sent to s3.. but this plugin don`t work with the additional websites.

How to enable Amazon s3 and cloud front plugin in site of the multisite wp network?

The plugin is network activated, and should work across all the sites in the network. What version of WP is the network running? Do you have the blogs.dir folder within wp-content?
