The previous answer was given too quickly and needs to be corrected. It is not possible to use the plugin without admin-ajax.php, because an Ajax request is fired after submitting a download form. This is done via admin-ajax.php.
Since many plugins use this file, it is good to think about whether it is useful to restrict it. Here you can find more information about this topic.
