I don’t understand the question? If you use page content and have your front page as the posts page then it would show you content in one column with text. Like this page: http://themes.kadencethemes.com/virtue/blog/

Currently, on my site in the previous post..I have a custom headline right below the slider images that reads “Check out the latest from our blog” and below that I have the “latest posts” lay out.
My question then is this…if I use “post content” rather than “latest posts” can I somehow have “Check out the latest from our blog” immediately below the slider images at the top and the start of the “post content”?
Not with the free theme. there is an option to add the “page title” but if you don’t have a static page that title would be “latest posts”.
Else you can alway use a child theme and edit the code to add whatever you like.

What code would I have to edit and in what file/folder in the child theme?

You would be editing the font-page.php file in the theme folder.
