As per your settings, location selection is OFF on cart and product page .
I don’t want the customer to select the location from front end.
make sure you have selected any location as default or at least one location for Auto order allocation.
Already did and the stock is deducted but the order note is missing.

by the way, your note from your screenshot is not a default woocommerece note
